Brandão Description An account of the resource Bovine coronavirus (BCoV) is a member of the group 2 of the Coronavirus (Nidovirales: Coronaviridae) and the causative agent of enteritis in both calves and adult bovine, as well as respiratory disease in calves. The present study aimed to develop a semi-nested RT-PCR for the detection of BCoV based on representative up-to-date sequences of the nucleocapsid gene, a conserved region of coronavirus genome. Three primers were designed, the first round with a 463bp and the second (semi-nested) with a 306bp predicted fragment. The analytical sensitivity was determined by 10-fold serial dilutions of the BCoV Kakegawa strain (HA titre: 256) in DEPC treated ultra-pure water, in fetal bovine serum (FBS) and in a BCoV-free fecal suspension, when positive results were found up to the 10-2, 10-3 and 10-7 dilutions, respectively, which suggests that the total amount of RNA in the sample influence the precipitation of pellets by the method of extraction used. When fecal samples was used, a large quantity of total RNA serves as carrier of BCoV RNA, demonstrating a high analytical sensitivity and lack of possible substances inhibiting the PCR. The final semi-nested RT-PCR protocol was applied to 25 fecal samples from adult cows, previously tested by a nested RT-PCR RdRp used as a reference test, resulting in 20 and 17 positives for the first and second tests, respectively, and a substantial agreement was found by kappa statistics (0.694). Los resultados muestran que los rotíferos cultivados en las salinidades de 35 ‰ y 25 ‰ presentan mayor ritmo de crecimiento poblacional y capacidad reproductiva que los cultivados en 10 y 7 ‰, durante los dos días de estudio. Asimismo, a 35 ‰ se corresponden los mayores valores de tasa de crecimiento (K: 1.66) y rendimiento (r: 129), seguido de 25 ‰. No obstante, en 25 ‰ se observó la mayor cantidad de rotíferos en etapa reproductiva (96 % de rotíferos con huevos y valor de índice reproductivo de 0.964). O tomate que apresenta produção e consumo mais difundidos no mundo, quer in natura ou processado, é altamente perecível, com perdas estimadas entre 25 a 50% da produção. Sendo assim, esta pesquisa objetivou a realização do processamento de tomates orgânicos desidratados em conservas obtidos a partir de dois diferentes equipamentos (desidratador e forno doméstico), com utilização de padrões para a pequena agroindústria e artesanais. Utilizaram-se três variedades de tomates orgânicos (Forty, Andrea e Perinha). Estes foram desidratados e em seguida foram processados na forma de conservas com uso de azeite de oliva como líquido de cobertura e condimentos orgânicos. Os resultados apresentaram produtos com umidade média que variou entre 46 a 63%, pH entre 4,07 e 4,31, cinzas entre 1,04 e 1,87%, proteínas entre 3,01 e 3,94%, lipídeos entre 19,77 e 28,17%, carboidratos entre 12,16 e 23,63% e VCT entre 241 e 348 calorias/100g. Untreated depression during pregnancy may have long-term effects on the mother and offspring. Therefore, delineating the effects of pregnancy on the mental health of reproductive-age women is crucial. This study aims to determine the risk for depressive symptoms in pregnant and non-pregnant women during COVID-19, and to identify its bio-psycho-social contributors. A total of 1114 pregnant and 256 non-pregnant women were recruited via social media in May 2020 to complete an online survey that included depression and anxiety questionnaires, as well as demographic, obstetric and COVID-19-related questionnaires. Pregnant women also completed the Pandemic-Related Pregnancy Stress Scale (PREPS). Pregnant women reported fewer depressive symptoms and were less concerned that they had COVID-19 than non-pregnant women. Among pregnant women, risk factors for depression included lower income, fewer children, unemployment, thinking that one has COVID-19, high-risk pregnancy, earlier gestational age, and increased pregnancy-related stress. Protective factors included increased partner support, healthy behaviors, and positive appraisal of the pregnancy. Thus, being pregnant is associated with reduced risk for depressive symptoms during the pandemic. Increased social support, engaging in health behaviors and positive appraisal may enhance resilience. Future studies of pregnant versus non-pregnant women could clarify the role of pregnancy during stressful events, and clarify aspects of susceptibility and resilience during pregnancy. Gray Description An account of the resource This case study entailed a Twitter content analysis to address the pandemic-delayed start to Major League Baseball (MLB) in the shortened 2020 season. This case study helps address the overarching objective to investigate how the sports world, especially fans, responded to MLB played during the 2020 COVID-19 pandemic. The methods investigated the common themes and determined who used predetermined Twitter hashtags. We recorded how many times external links, photos, emojis, and the 30 MLB teams were mentioned in the 779 tweets obtained during 39 days of data retrieval. Results showed that the most common category of tweeted content concerned news reports. Comparable numbers of positive and negative responses to the start of the MLB season were recognized, with a fraction of tweets highlighting COVID-19 impacts on health and modification of play (e.g., cardboard fans). The majority of Twitter users were from media and layperson categories. More inferred males tweeted using the selected hashtags. In exploratory analyses, results indicated that 50.2% of the sample included a link or a photo, and 2.2% of the sample used an emoji. The three most mentioned teams were the Cardinals (N = 51), Marlins (N = 49), and the Yankees (N = 48). The results confirmed the value of social media analysis as a research approach and revealed patterns emerging during a unique pandemic sports and media era. Standard single population models typically suffer from two major drawbacks: on the one hand, they use a large number of parameters compared to the sample size and, on the other hand, model choice is still often based on in-sample criterion, such as the Bayes information criterion (BIC), and therefore not on the ability to predict. In this paper, we develop a model based on a decomposition of the mortality surface into a polynomial basis. Then, we show how regularization techniques and cross-validation can be used to obtain a parsimonious and coherent predictive model for mortality forecasting. We analyze how COVID-19-type effects can affect predictions in our approach and in the classical one. In particular, death rates forecasts tend to be more robust compared to models with a cohort effect, and the regularized model outperforms the so-called P-spline model in terms of prediction and stability.
