                <text>Abstract Background Despite the passing of more than a year since the first outbreak of Severe Acute Respiratory Syndrome (SARS), efficient counter-measures are still few and many believe that reappearance of SARS, or a similar disease caused by a coronavirus, is not unlikely. For other virus families like the picornaviruses it is known that pathology is related to proteolytic cleavage of host proteins by viral proteinases. Furthermore, several studies indicate that virus proliferation can be arrested using specific proteinase inhibitors supporting the belief that proteinases are indeed important during infection. Prompted by this, we set out to analyse and predict cleavage by the coronavirus main proteinase using computational methods. Results We retrieved sequence data on seven fully sequenced coronaviruses and identified the main 3CL proteinase cleavage sites in polyproteins using alignments. A neural network was trained to recognise the cleavage sites in the genomes obtaining a sensitivity of 87.0% and a specificity of 99.0%. Several proteins known to be cleaved by other viruses were submitted to prediction as well as proteins suspected relevant in coronavirus pathology. Cleavage sites were predicted in proteins such as the cystic fibrosis transmembrane conductance regulator (CFTR), transcription factors CREB-RP and OCT-1, and components of the ubiquitin pathway. Conclusions Our prediction method NetCorona predicts coronavirus cleavage sites with high specificity and several potential cleavage candidates were identified which might be important to elucidate coronavirus pathology. Furthermore, the method might assist in design of proteinase inhibitors for treatment of SARS and possible future diseases caused by coronaviruses. O experimento foi conduzido em casa de vegetação e contou com seis tratamentos de sementes, constituídos por: bioestimulante (T1), bioestimulante + fipronil (T2), bioestimulante + fipronil + piraclostrobina + tiofanato-metílico (T3), bioestimulante + abamectina (T4), bioestimulante + thiamethoxam (T5) e bioestimulante + imidacloprido + tiodicarbe (T6). As avaliações ocorreram 21 dias após a semeadura da soja, no estádio fenológico V4-V5. Foi avaliado: massa seca da parte aérea, massa seca da raiz, diâmetro do caule e altura de planta. A adição de inseticidas no tratamento de sementes de soja com bioestimulante, nas condições em que o experimento foi realizado, gerou acréscimos para a massa seca das raízes. Destacando-se o ingrediente ativo Thiamethoxam. Os inseticidas não proporcionaram incrementos na massa seca da parte aérea e na altura de plantas de soja. O diâmetro do caule apresentou respostas variáveis de acordo com o inseticida utilizado.</text>

                <text>Uno de los aspectos menos abordados en el ámbito académico de las Relaciones Internacionales, y más específicamente en el de la cooperación internacional, es el referente al papel que la ayuda internacional puede tener en el fortalecimiento de los procesos de integración regional. Este aspecto ha sido poco trabajado en las universidades de los países que son Estados – Parte del Mercado Común del Sur (MERCOSUR), a pesar de que el apoyo principalmente de la Unión Europea (UE) y de algunas agencias multilaterales y bilaterales haya sido relevante para el surgimiento y consolidación de algunas instituciones propias (Secretaría Administrativa, Parlamento, Comisión de Representantes Permanentes), para la puesta en marcha de algunas iniciativas integracionistas en campos como el medio ambiente, la agricultura familiar o el género, o aún, para el fortalecimiento de las redes de la sociedad civil. Este artículo pretende incentivar la discusión y el debate sobre las posibilidades de la cooperación internacional en el apoyo al MERCOSUR. El texto se divide en tres apartados y unas conclusiones. En la primera parte se presentan las estructuras institucionales y las disposiciones de los órganos de integración relativas a la cooperación internacional. En la segunda parte se describe y analiza la cooperación recibida por MERCOSUR en tres categorías: agencias bilaterales, UE y Organismos Multilaterales. En la tercera parte se presenta el incipiente programa subregional de cooperación con el MERCOSUR, de la Agencia Española de Cooperación Internacional para el Desarrollo (AECID). Por último, se extraen unas conclusiones sobre el estado de la cooperación recibida por el MERCOSUR.</text>

                <text>Sea biscuits and sand dollars diverged from other irregular echinoids approximately 55 million years ago and rapidly dispersed to oceans worldwide. A series of morphological changes were associated with the occupation of sand beds such as flattening of the body, shortening of primary spines, multiplication of podia, and retention of the lantern of Aristotle into adulthood. To investigate the developmental basis of such morphological changes we documented the ontogeny of Clypeaster subdepressus. We obtained gametes from adult specimens by KCl injection and raised the embryos at 26 degrees C. Ciliated blastulae hatched 7.5 h after sperm entry. During gastrulation the archenteron elongated continuously while ectodermal red-pigmented cells migrated synchronously to the apical plate. Pluteus larvae began to feed in 3 d and were 20 d old at metamorphosis; starved larvae died 17 d after fertilization. Postlarval juveniles had neither mouth nor anus nor plates on the aboral side, except for the remnants of larval spicules, but their bilateral symmetry became evident after the resorption of larval tissues. Ossicles of the lantern were present and organized in 5 groups. Each group had 1 tooth, 2 demipyramids, and 2 epiphyses with a rotula in between. Early appendages consisted of 15 spines, 15 podia (2 types), and 5 sphaeridia. Podial types were distributed in accordance to Lovén's rule and the first podium of each ambulacrum was not encircled by the skeleton. Seven days after metamorphosis juveniles began to feed by rasping sand grains with the lantern. Juveniles survived in laboratory cultures for 9 months and died with wide, a single open sphaeridium per ambulacrum, aboral anus, and no differentiated food grooves or petaloids. Tracking the morphogenesis of early juveniles is a necessary step to elucidate the developmental mechanisms of echinoid growth and important groundwork to clarify homologies between irregular urchins.</text>

                <text>Abstract Background Stereotypic cleavage patterns play a crucial role in cell fate determination by precisely positioning early embryonic blastomeres. Although misplaced cell divisions can alter blastomere fates and cause embryonic defects, cleavage patterns have been modified several times during animal evolution. However, it remains unclear how evolutionary changes in cleavage impact the specification of blastomere fates. Here, we analyze the transition from spiral cleavage – a stereotypic pattern remarkably conserved in many protostomes – to a biradial cleavage pattern, which occurred during the evolution of bryozoans. Results Using 3D-live imaging time-lapse microscopy (4D-microscopy), we characterize the cell lineage, MAPK signaling, and the expression of 16 developmental genes in the bryozoan Membranipora membranacea. We found that the molecular identity and the fates of early bryozoan blastomeres are similar to the putative homologous blastomeres in spiral-cleaving embryos. Conclusions Our work suggests that bryozoans have retained traits of spiral development, such as the early embryonic fate map, despite the evolution of a novel cleavage geometry. These findings provide additional support that stereotypic cleavage patterns can be modified during evolution without major changes to the molecular identity and fate of embryonic blastomeres.</text>

                <text>Staphylococcus aureus is a major human pathogen causing a wide range of nosocomial infections including pulmonary, urinary, and skin infections. Notably, the emergence of bacterial strains resistant to conventional antibiotics has prompted researchers to find new compounds capable of killing these pathogens. Nature is undoubtedly an invaluable source of bioactive molecules characterized by an ample chemical diversity. They can act as unique platform providing new scaffolds for further chemical modifications in order to obtain compounds with optimized biological activity. A class of natural compounds with a variety of biological activities is represented by alkaloids, important secondary metabolites produced by a large number of organisms including bacteria, fungi, plants, and animals. In this work, starting from the screening of 39 alkaloids retrieved from a unique in-house library, we identified a heterodimer β-carboline alkaloid, nigritanine, with a potent anti-Staphylococcus action. Nigritanine, isolated from Strychnos nigritana, was characterized for its antimicrobial activity against a reference and three clinical isolates of S. aureus. Its potential cytotoxicity was also evaluated at short and long term against mammalian red blood cells and human keratinocytes, respectively. Nigritanine showed a remarkable antimicrobial activity (minimum inhibitory concentration of 128 µM) without being toxic in vitro to both tested cells. The analysis of the antibacterial activity related to the nigritanine scaffold furnished new insights in the structure–activity relationships (SARs) of β-carboline, confirming that dimerization improves its antibacterial activity. Taking into account these interesting results, nigritanine can be considered as a promising candidate for the development of new antimicrobial molecules for the treatment of S. aureus-induced infections.</text>

                <text>Cellular and viral S-adenosylmethionine-dependent methyltransferases are involved in many regulated processes such as metabolism, detoxification, signal transduction, chromatin remodeling, nucleic acid processing, and mRNA capping. The Severe Acute Respiratory Syndrome coronavirus nsp16 protein is a S-adenosylmethionine-dependent (nucleoside-2'-O)-methyltransferase only active in the presence of its activating partner nsp10. We report the nsp10/nsp16 complex structure at 2.0 Å resolution, which shows nsp10 bound to nsp16 through a ∼930 Å² surface area in nsp10. Functional assays identify key residues involved in nsp10/nsp16 association, and in RNA binding or catalysis, the latter likely through a SN2-like mechanism. We present two other crystal structures, the inhibitor Sinefungin bound in the S-adenosylmethionine binding pocket and the tighter complex nsp10(Y96F)/nsp16, providing the first structural insight into the regulation of RNA capping enzymes in +RNA viruses.</text>

                <text>Con la salida de la convertibilidad, la actividad porcina en Argentina se transformó en una producción rentable. Analizando el período 2002-2015, se puede afirmar que tuvo un vuelco cualitativo en la forma de producir carne. Esto demanda manejo y reutilización de los volúmenes de efluentes que genera la producción intensiva. Sin un tratamiento adecuado, pueden resultar en una fuente de contaminación y deterioro del medio ambiente. Los objetivos del presente trabajo final de graduación fueron delinear instalaciones para un eficiente tratamiento biológico de purines en biogás, estimar las conversiones del biogás producido en energía térmica y eléctrica, determinar el uso consultivo de la cogeneración térmica y eléctrica en el sistema de producción y evaluar el aprovechamiento del agua tratada en el proceso biológico para la recirculación en las fosas de los sitios de producción. Para esto se realizó una revisión bibliográfica intensiva, se procedió a calcular DBO5 y DQO por cada 100 kilos de PV. Se efectuó el cálculo y dimensionamiento de fosas de recolección de purines del sistema y tándem lagunar para vaciado bimestral, en un establecimiento en confinamiento de 1500 madres. Finalmente, se observó que, aunque la cantidad de biogás generada en un establecimiento porcino confinado no logre cogenerar la cantidad de energía térmica y eléctrica que demandada, puede contribuir a cubrir cierta parte y a su vez aportar biofertilizantes no contaminantes ya que, durante la digestión anaeróbica en los biodigestores, la DBO5 disminuye un 90%.</text>

                <text>El objetivo de la investigación fue evaluar la sustentabilidad de los agroecosistemas cafetaleros conducidos por los agricultores familiares del distrito de Vitoc, Región Junín. Se eligieron seis localidades del distrito y dentro de cada localidad, en forma proporcional, las propiedades de los agricultores familiares. Se evaluaron las dimensiones económicas (rentabilidad, ingreso neto mensual y riesgo económico), ambiental (conservación de la vida del suelo, riego de erosión y manejo de biodiversidad) y social (satisfacción de las necesidades básicas, integración social y conciencia ecológica) y el Índice de Sustentabilidad General. Asimismo, para considerar que el agroecosistema es sustentable, se consideró que el indicador debe ser mayor a 2 en todas las dimensiones y en el índice general. Los resultados muestran que los indicadores obtenidos para las dimensiones económica, ambiental, social y el índice general son favorables y orientan hacia una agricultura sustentable. Sin embargo, presenta problemas con la diversificación de los cultivos para la venta, protección del suelo y la cobertura sanitaria, las que pueden ser considerados como una debilidad del agroecosistema. Se concluye que, para las condiciones del estudio, los agroecosistemas cafetaleros conducidos por los agricultores familiares son sustentables.</text>
