BACKGROUND:Entry of enveloped viruses into host cells requires the activation of viral envelope glycoproteins through cleavage by either intracellular or extracellular proteases. In order to gain insight into the molecular basis of protease cleavage and its impact on the efficiency of viral entry, we investigated the susceptibility of a recombinant native full-length S-protein trimer (triSpike) of the severe acute respiratory syndrome coronavirus (SARS-CoV) to cleavage by various airway proteases. METHODOLOGY/PRINCIPAL FINDINGS:PURIFIED TRISPIKE PROTEINS WERE READILY CLEAVED IN VITRO BY THREE DIFFERENT AIRWAY PROTEASES: trypsin, plasmin and TMPRSS11a. High Performance Liquid Chromatography (HPLC) and amino acid sequencing analyses identified two arginine residues (R667 and R797) as potential protease cleavage site(s). The effect of protease-dependent enhancement of SARS-CoV infection was demonstrated with ACE2 expressing human bronchial epithelial cells 16HBE. Airway proteases regulate the infectivity of SARS-CoV in a fashion dependent on previous receptor binding. The role of arginine residues was further shown with mutant constructs (R667A, R797A or R797AR667A). Mutation of R667 or R797 did not affect the expression of S-protein but resulted in a differential efficacy of pseudotyping into SARS-CoVpp. The R667A SARS-CoVpp mutant exhibited a lack of virus entry enhancement following protease treatment. CONCLUSIONS/SIGNIFICANCE:These results suggest that SARS S-protein is susceptible to airway protease cleavage and, furthermore, that protease mediated enhancement of virus entry depends on specific conformation of SARS S-protein upon ACE2 binding. These data have direct implications for the cell entry mechanism of SARS-CoV along the respiratory system and, furthermore expand the possibility of identifying potential therapeutic agents against SARS-CoV.

A new detailed aqueous phase mechanism named the Cloud Explicit Physico-chemical Scheme (CLEPS 1.0) is proposed to describe the oxidation of water soluble organic compounds resulting from isoprene oxidation. It is based on structure activity relationships (SARs) which provide global rate constants together with branching ratios for HO⋅ abstraction and addition on atmospheric organic compounds. The GROMHE SAR allows the evaluation of Henry's law constants for undocumented organic compounds. This new aqueous phase mechanism is coupled with the MCM v3.3.1 gas phase mechanism through a mass transfer scheme between gas phase and aqueous phase. The resulting multiphase mechanism has then been implemented in a model based on the Dynamically Simple Model for Atmospheric Chemical Complexity (DSMACC) using the Kinetic PreProcessor (KPP) that can serve to analyze data from cloud chamber experiments and field campaigns. The simulation of permanent cloud under low-NOx conditions describes the formation of oxidized monoacids and diacids in the aqueous phase as well as a significant influence on the gas phase chemistry and composition and shows that the aqueous phase reactivity leads to an efficient fragmentation and functionalization of organic compounds.

Para determinar la relación entre el clima y la variabilidad de la temperatura sub-superficial, se analizaron los registros de la temperatura sub-superficial del mar de nueve estaciones localizadas en la Bahía. El análisis permitió caracterizar su ciclo anual y explorar su relación con fuentes de variabilidad climática que influencian el clima regional para el periodo 1998-2010. Los resultados se contextualizaron usando además los datos de una estación meteorológica automática que funcionó en la bahía junto con el registro de la temperatura superficial del mar para una rejilla que cubre la región de 9-11°N, 85-87°W, para el periodo 1854-2011. Debido a la tendencia positiva encontrada en la región para la temperatura superficial del mar, se concluye que los resultados mostrados asociados a los ciclos anuales y diarios en Bahía Culebra deben ser interpretados bajo un escenario de cambio climático, asociado a un calentamiento ocurrido desde 1854, además coherente con lo observado globalmente, y que su variabilidad climática está influenciada no sólo por aquella ligada a la de El Niño-Oscilación del Sur, en el Pacífico Ecuatorial, sino también por influencias de tipo atmosférico relacionadas con la variabilidad en el Océano Atlántico, debido a que los eventos cálidos (fríos) en Bahía Culebra tienden a ocurrir en concordancia con anomalías positivas y negativas (negativas y positivas) de los índices Niño 3.4 y OAN, respectivamente.

Bahía Culebra, Golfo de Papagayo, Costa Rica is a seasonal upwelling area. To determine the relationship of climate and the subsurface temperature variability at Bahía Culebra, we analyzed nine records of sea subsurface temperature from the Bay, continuously recorded from 1998 to 2010. The analysis characterized the annual cycle and explored the influence of different climate variability sources on the subsurface sea temperature and air temperature recorded in Bahía Culebra. Data from an automatic meteorological station in the bay were studied, obtaining the annual and daily cycle for air surface temperature and wind speed. Sea surface temperature (SST) trend from 1854 to 2011 was calculated from reanalysis for the region that coverts 9-11°N, 85-87°W. Because of the positive SST trend identified in this region, results showed that annual and daily cycles in Bahía Culebra should be studied under a warming scenario since 1854, that is coherent with the global warming results and its climate variability is influenced by El Niño-Southern Oscillation (ENSO) in the Equatorial Pacific and by atmospheric forcing triggered by climate variability with Atlantic Ocean origin, because warm (cold) events in Bahía Culebra tend to occur in concordance with positive & negative (negative & positive) anomalies in Niño 3.4 (NAO) index.

La climatología es la rama que desarrolla en mayor grado el principio rector de la geografía: el estudio de las relaciones entre el medio y el ser humano, así como de sus diferentes manifestaciones sobre la superficie terrestre. La climatología española compite en la actualidad en igualdad de condiciones con el análisis climático internacional. Ha incluido nuevos planteamientos teóricos y métodos de trabajo que proporcionan resultados publicados en revistas y en monografías de impacto. Uno de los temas principales de la investigación española en dicho ámbito es el cambio climático actual y los efectos que ejerce en los elementos atmosféricos. El litoral mediterráneo es un buen laboratorio para profundizar en esta cuestión, puesto que ya manifiesta consecuencias que, si mantienen su tendencia actual, pueden comportar efectos en las actividades económicas y en la propia ocupación del territorio. En el presente artículo se propone una agenda de trabajo para que, en este contexto, la climatología española se convierta en la rama protagonista de la investigación y la docencia geográfica española en los próximos años.

The upper treeline provides a unique opportunity for documenting changes in forest dynamics in the context of current environmental fluctuations. Documenting the response of forests to present environmental changes will be very useful in developing and validating vegetation models that predict plant responses to global changes resulting from human activities. It is generally suggested that the expansion of the upper treeline is limited by cold temperatures, so warmer conditions are expected to induce an advance of forests into alpine vegetation. Significant changes in temperature and precipitation have already been documented in regional studies along the Patagonian Andes during the last decades. Across Patagonia, there are strong relationships between changes in climate and variations in the Southern Annular Mode (SAM). The atmospheric circulation indexes, act as climate packages summarizing climatic information since they combine changes in environmental variables. In order to complement previous studies in the northern Patagonia treeline, we have conducted a this study in the southern Patagonian Andes to: (1) characterize the patterns of Nothofagus pumilio establishment in the upper treeline along a precipitation gradient, (2) to establish the relationship between variations in regional climate and tree establishment and (3) to determine the influence of continental and hemispheric-scale climatic forcing on tree establishment at regional scale. Our studies suggest that the location of the upper treeline of the N. pumilio forest in the Patagonian Andes is sensitive to changes in climate variations. The temperature increment has facilitated the tree recruitment, however, the rate of seedling establishment appears to be more strongly modulated by the interaction between temperature increase and variations in precipitation. The expansion of the upper treeline has been limited to 5–10m from the abrupt treeline. This comparatively reduced advance of the forest in the alpine grassland is regulated by other biotic and abiotic factors rather than the temperature increase. While the recent expansion of the upper treeline in northern Patagonia was regulated by temperature and precipitation variations associated with the different phases of the Pacific Decadal Oscillation, the establishment above the upper treeline in the southern Patagonian Andes was facilitated by climatic variations induced by the positive trend of the SAM during the last decades.

This article analyzes the possible impacts of the climate change (CC) and its adaptation measures on the maize trade surplus/deficit, on the production, and on the real income of households established in rural Mexico. The analysis is based on an Applied and Microeconomic General Equilibrium Model and on the estimates of the direct effects of CC on maize yields obtained from the related literature. The results show that climate change will reduce the real rural income by 6.23% and will affect corn production to the extent that the area will change from being a surplus to a deficit. In addition, it is found that while the adaptation measures analyzed in the paper will help to reduce the negative effects of the phenomenon, they will not completely eliminate them. Adaptación al cambio climático y sus efectos en la economía de los hogares rurales mexicanos productores de alimentos ABSTRACT En este artículo, se analizan los posibles impactos del Cambio Climático (CC) y de sus medidas de adaptación en el superávit/déficit comercial del maíz, en la producción y los ingresos reales de los hogares establecidos en la zona rural de México, a partir de un Modelo de Equilibrio General Aplicado y Microeconómico, y de estimaciones directas de los efectos del CC en los rendimientos del maíz obtenidas de la literatura relacionada. Los resultados encontrados muestran que el cambio climático reducirá el ingreso real rural en 6.23% y que afectará, en mayor medida, a la producción de maíz, al grado que la zona pasará de ser superavitaria a deficitaria. Además, se obtuvo que, si bien las medidas de adaptación analizadas ayudarán a reducir los efectos negativos del fenómeno, estas no los eliminarán totalmente.

This research has the objective of analyzing whether the public policies that today define the rules for productive development in agriculture can mitigate the effects related to global warming and if these are sufficient to prevent the pressing of climate change over the difficulties already existing in the agricultural sector or, to prevent the emergence of new risk factors or threats that affect the country’s food security. It also seeks to propose some recommendations for the design of public policies to minimize the risks and threats related to the adaptation of agricultural production to climate change and, therefore, to ensure physical, social and economic access to sufficient food and nutritious foods that meet the needs of the national population and mitigate its effects on the multidimensional sphere of the security. In this sense, we try to answer the following question: What challenges does climate change pose to Chilean agriculture in terms of food security?
