There are several diagnostic techniques used to detect BCoV in calf fecal samples, but virus isolation still has advantages for antigenic and genomic characterization. This study describes the isolation in HRT-18 cells and molecular characterization of Brazilian BCoV wild-type strains. Three fecal samples from diarrheic 30 day-old calves were inoculated in HRT-18 cell monolayers, which were then evaluated for HA titers and tested using semi-nested PCR followed by RFLP and sequencing. Two samples were successfully isolated and presented HA titers of 16 and 32 units per 25 mL. The results were confirmed using semi-nested PCR and RFLP. Molecular analyses identified a cell culture-adapted strain and a wild-type strain that were genetically similar (99%) to each other, but more distinct than BCoV strains circulating in other countries, even in the conserved N gene.O coronavírus bovino (BCoV) pode causar diarreia aguda em bezerros recém-nascidos, ocasionando consideráveis perdas econômicas para a pecuária bovina. Porém, o isolamento do BCoV em cultivo celular apresenta a vantagem de possibilitar a caracterização antigênica e molecular da estirpe viral. O presente estudo descreve o isolamento em células HRT-18, e a caracterização molecular de estirpes brasileiras do BCoV. Três amostras de fezes diarreicas de bezerros com 30 dias de idade foram inoculadas em culturas de células HRT-18. Os isolados foram avaliados por hemaglutinação (HA) e por uma semi-nested PCR seguida de RFLP e sequenciamento. De los tratamientos que conforman el ensayo sólo se seleccionaron cuatro secuencias de cultivos agrícolas: soja-soja (S-S), soja-trigo/soja (S-T/S), maíz-soja (M-S) y maíz-trigo/soja (M-T/S). Sobre las muestras de suelo se determinaron parámetros biológicos, como respiración microbiana y actividades enzimáticas, y parámetros químicos como contenido de materia orgánica y macronutrientes. Arteriviruses and coronaviruses, distantly related members of the nidovirus order, employ a unique mechanism of discontinuous minus-strand RNA synthesis to generate subgenome-length templates for the synthesis of a nested set of sg mRNAs. Non-structural protein 1 (nsp1) of the arterivirus equine arteritis virus (EAV), a multifunctional regulator of viral RNA synthesis and virion biogenesis, was previously implicated in controlling the balance between genome replication and sg mRNA synthesis. Here, we employed reverse and forward genetics to gain insight into the multiple regulatory roles of nsp1. Our analysis revealed that the relative abundance of viral mRNAs is tightly controlled by an intricate network of interactions involving all nsp1 subdomains. Distinct nsp1 mutations affected the quantitative balance among viral mRNA species, and our data implicate nsp1 in controlling the accumulation of full-length and subgenome-length minus-strand templates for viral mRNA synthesis. The moderate differential changes in viral mRNA abundance of nsp1 mutants resulted in similarly altered viral protein levels, but progeny virus yields were greatly reduced. Pseudorevertant analysis provided compelling genetic evidence that balanced EAV mRNA accumulation is critical for efficient virus production. This first report on protein-mediated, mRNA-specific control of nidovirus RNA synthesis reveals the existence of an integral control mechanism to fine-tune replication, sg mRNA synthesis, and virus production, and establishes a major role for nsp1 in coordinating the arterivirus replicative cycle. Uno de los procesos más prometedores para producir etanol a partir de biomasa lignocelulósica es la sacarificación y fermentación simultáneas (SSF). En otros estudios realizados, con experimentos SSF, se ha evaluado la conversión de glucano en etanol a partir de la cáscara de Jatropha pretratada en medio ácido o alcalino pero no a través de procesos hidrotérmicos. En el presente estudio, la cáscara de Jatropha curcas fue pretratada hidrotérmicamente con agua caliente a 200 °C por 15 min, y ensayada en experimentos de SSF con un coctel enzimático (Cellic CTec 2) y levadura comercial (Saccharomyces cerevisiae). Diferentes cargas enzimáticas, medios estériles y tamaños de ensayo fueron usados para determinar su influencia en el rendimiento de etanol. Los resultados mostraron que, con 5% p/p de sólidos, 15 UPF/g biomasa seca, y 1mg/ml de levadura se alcanza el máximo rendimiento de etanol (0,160- 0,177 g etanol/ g biomasa seca) y conversión de celulosa (57,71- 63,90%). A través de esta investigación se propone una alternativa para el uso integral del cultivo de Jatropha curcas dentro de una economía circular. Método: se usó muestreo sistemático, con sitios cada 100 m s. n. m. entre los 2900 y 3600 m s. n. m. Con el programa sas se hicieron pruebas de normalidad, tablas de frecuencia y análisis de varianza para analizar la relación entre altitud, vigor y regeneración de Abies religiosa (Kunth) Schltdl. et Cham. Resultados: hay relación entre la altitud, el vigor y la densidad arbórea. La regeneración es nula en las cotas más bajas e incrementa con la altitud hasta los 3 400 m s. n. m. y no muestra gran reducción más arriba. La densidad de los estratos A (dap ≥12.5 cm) y B (dap 1 m de altura) incrementa conforme a la altitud; las densidades más altas se registran en la cota 3 400 m s. n. m. Limitaciones: se requiere investigación de otros factores que intervienen en la distribución de la especie. Principales hallazgos: la regeneración tiene más éxito en zonas medias y altas, y la condición del arbolado tiende a ser mejor ahí también. Se evidencia que el oyamel migra a zonas más elevadas, debido al cambio climático global.
