We describe the case of a young female patient admitted to our emergency department during the Italian COVID-19 epidemic, for fever and dry cough associated with symptoms of gastric reflux over the previous 5 days. Lung ultrasound showed diffuse bilateral B lines with irregular pleural thickening, and consolidation with air bronchogram and slight pleural effusion in the lower left lobe. Chest HRCT and abdominal CT scanning with contrast revealed diaphragmatic rupture with gastric perforation, and atelectasis of the left pulmonary lobe with unilateral pleural effusion, diffuse ground-glass opacities and multiple small consolidations in both lobes. A nasopharyngeal swab for 2019-nCoV was positive. A diagnosis of diaphragmatic rupture and gastric perforation in COVID-19 pneumonia was made. The patient was immediately hospitalized and surgically treated. Treatment for COVID-19 and empiric antibiotic therapy were promptly started.

Coronavirus disease 2019 (COVID-19) is a new infectious disease that spreads very rapidly and therefore, WHO has declared it as a global pandemic disease. The main clinical symptoms found in COVID-19 patients are cough and fever; however, in some cases, diarrhea can be one of the early symptoms. The present case report describes a patient who came with a complaint of diarrhea without fever and she was later confirmed to be positive for COVID-19 during hospitalization. The presence of unspecified initial symptoms calls for greater vigilance from health workers in establishing diagnosis patients with COVID-19.

5’,8-Cyclo-2’-deoxyadenosine (cdA), in the 5’R and 5’Sdiastereomeric forms, are typical non strand-break oxidative DNA lesions, induced by hydroxyl radicals, with emerging importance as a molecular marker. These lesions are exclusively repaired by the nucleotide excision repair (NER) mechanism with a low efficiency, thus readily accumulating in the genome. Poly(ADP-ribose) polymerase1 (PARP1) acts as an early responder to DNA damage and plays a key role as a nick sensor in the maintenance of the integrity of the genome by recognizing nicked DNA. So far, it was unknown whether the two diastereomeric cdA lesions could induce specific PARP1 binding. Here, we provide the first evidence of PARP1 to selectively recognize the diastereomeric lesions of 5’S-cdA and 5’R-cdA in vitro as compared to deoxyadenosine in model DNA substrates (23-mers) by using circular dichroism, fluorescence spectroscopy, immunoblotting analysis, and gel mobility shift assay. Several features of the recognition of the damaged and undamaged oligonucleotides by PARP1 were characterized. Remarkably, PARP1 exhibits different affinities in binding to a double strand (ds) oligonucleotide, which incorporates cdA lesions in R and S diastereomeric form. In particular, PARP1 proved to bind oligonucleotides, including a 5’S-cdA, with a higher affinity constant for the 5’S lesion in a model of ds DNA than 5’R-cdA, showing different recognition patterns, also compared with undamaged dA. This new finding highlights the ability of PARP1 to recognize and differentiate the distorted DNA backbone in a biomimetic system caused by different diastereomeric forms of a cdA lesion.

The seasonal variation of diatoms in the inshore waters off Paraná State, Southern Brazil was investigated to analyse their temporal dynamics and to detect the main environmental constraints of the planktonic community. Biomass peaks occurred from May to August and from December to March. Among the microplanktonic diatoms, Cerataulina pelagica, Chaetoceros spp., Dactyliosolen fragilissimus, Guinardia striata, Lauderia annulata, Leptocylindrus spp., Pseudo-nitzschia cf. delicatissima, P. australis, Rhizosolenia spp., Skeletonema costatum and Thalassionema nitzschioides were dominant species. Nanoplanktonic diatoms were dominated by Naviculaceae, Nitzschia spp., Thalassiosira spp. and Chaetoceros cf. tenuissimus. Concentrations of most of the species decreased during and just after the blooms of Phaeocystis pouchetii in September and of Coscinodiscus wailesii in April. Based on cluster analysis and interpretations of the environmental parameters monitored, six diatom associations were discerned. Four main environmental factors were attributed as the determinants for the grouping: (a) the stronger influence of the tropical oligotrophic waters of the Brazil Current in spring/summer, (b) the alternation between dry and rainy (nutrient-richer) seasons, (c) the influence of subantarctic waters mixed with coastal ones in fall/winter, bringing cold species and promoting the growth of autochthonous species, and (d) the blooms of nanoplanktonic (Phaeocystis) and microplanktonic (C. wailesii) species.A variação sazonal das diatomáceas em águas neríticas do estado do Paraná, Sul do Brasil, foi investigada para analisar sua dinâmica temporal e detectar as principais forçantes ambientais da comunidade planctônica. Picos de biomassa ocorreram de Maio a Agosto e de Dezembro a Março. As espécies dominantes foram Cerataulina pelagica, Chaetoceros spp., Dactyliosolen fragilissimus, Guinardia striata, Lauderia annulata, Leptocylindrus spp., Pseudo-nitzschia cf. delicatissima, P. australis, Rhizosolenia spp., Skeletonema costatum e Thalassionema nitzschioides. Representantes de Naviculaceae, Nitzschia spp., Thalassiosira spp. e Chaetoceros cf. tenuissimus dominaram as diatomáceas nanoplanctônicas. As concentrações da maioria das espécies decresceram durante e logo após as florações de Phaeocystis pouchetii em Setembro, e de Coscinodiscus wailesii em Abril. Com base na análise de cluster e interpretações dos parâmetros ambientais monitorados, seis associações de diatomáceas foram discernidas. Quatro fatores ambientais principais foram atribuídos como determinantes para o grupamento: (a) influência mais forte de águas oligotróficas da Corrente do Brasil na Primavera/Verão, (b) alternância entre as estações chuvosa (rica em nutrientes) e seca, (c) influência de águas subantárticas enriquecidas misturadas às costeiras no Outono e Inverno, trazendo espécies de águas frias, além de promover o crescimento de autóctones, e (d) florações de espécies nanoplanctônicas (Phaeocystis) e microplanctônicas (C. wailesii).

The association of wild grasses with diazotrophic bacteria in Brazilian biomes is poorly understood. The isolation and characterization of bacteria associated with wild grasses can contribute to understand the diazotrophic ecology as well as to identify bacteria with biotechnological applications. In this study, we isolated and characterized diazotrophic bacterial isolates from Oryza glumaepatula collected in Cerrado and Forest areas of the Amazon in Roraima State, Brazil. Healthy O. glumepatula plants were collected at five sampling sites at Forest and seven at Cerrado, respectively. The plants were collected at the Cerrado areas in September 2008 while the Forest plants were collected in June/2008 and April/2009. The plants and the soil adhering to the roots were transferred to pots and grown for 35 days in greenhouse conditions. During the harvest, the shoots and the roots were crushed separately in a saline solution; the suspension was diluted serially and inoculated in Petri dishes containing Dyg’s medium. All distinct bacterial colonies were purified in the same medium. The diazotrophic capacity of each bacterium in microaerophilic conditions was assessed in semisolid BMGM medium. In addition, the pellicles forming bacterial isolates were also evaluated by PCR amplification for nifH gene. The diversity of nifH+ bacteria was analyzed by Box-PCR fingerprinting. For selected strains, the growth promoting capacity of O. sativa as a model plant was also evaluated. A total of 992 bacterial isolates were obtained. Fifty- one bacteria were able to form pellicles in the semisolid medium and 38 also positively amplified the 360bp nifH gene fragment. Among the 38 nifH+ isolates, 24 were obtained from the shoots, while 14 originated from the roots. The Box-PCR profiles showed that the bacterial isolates obtained in this study presented a low similarity with the reference strains belonging to the Herbaspirillum, Azospirillum and Burkholderia genus. The growth- promoting ability was confirmed for at least five isolates. For these bacteria, the root and shoot growing results showed higher increases when compared to those observed in plants inoculated with the evaluated reference strains. These results indicate that O. glumaepatula is colonized by a high diverse diazotrophic community in the Brazilian Amazon. Further investigations are now being carried out to determine the taxonomic positions of these isolates and their growth promoting mechanisms.La asociación de gramíneas silvestres con bacterias diazotróficas en los biomas brasileños es poco conocida. El aislamiento y caracterización de las bacterias asociadas con gramíneas silvestres puede contribuir a entender la ecología de las diazotróficas y bacterias con aplicaciones biotecnológicas. En este estudio, caracterizamos aislamientos bacterianos de diazotróficas de Oryza glumaepatula recolectadas en Cerrado y zonas boscosas de la Amazonía en el estado de Roraima, Brasil. Plantas sanas de O. glumepatula fueron recolectadas en cinco zonas boscosas y siete en Cerrado. Las plantas de Cerrado fueron recolectadas en septiembre 2008, mientras que las del bosque en Junio 2008 y Abril 2009. Las plantas y el suelo adherido a las raíces se transfirieron a macetas y se cultivaron durante 35 días en condiciones de invernadero. Durante la cosecha, los brotes y las raíces se trituraron por separado en una solución salina, la suspensión se diluyó en serie y se inocularon en placas Petri que contenían medio Dyg. Todas las colonias de bacterias se purificaron en el mismo medio. Se evaluó la capacidad diazotrófica de cada bacteria en condiciones microaerofílicas en medio semisólido BMGM. Además, los aislamientos bacterianos que formaron películas se evaluaron también mediante amplificación por PCR para el gen nifH. La diversidad de bacterias nifH+ se analizó por Huella Genética utilizando la Reacción en Cadena de la Polimerasa. Para las cepas seleccionadas, la capacidad de promover el crecimiento de O. sativa como modelo de planta también se evaluó. Se obtuvo un total de 992 cepas bacterianas. Cincuenta y un bacterias fueron capaces de formar películas en el medio semisólido y 38 amplificaron positivamente el fragmento 360bp del gen nifH. De los 38 aislamientos de nifH+, 24 fueron obtenidos de los brotes, mientras que 14 se originaron a partir de las raíces. Los perfiles de PCR-Box mostraron que los aislamientos bacterianos obtenidos en este estudio presentaron una baja similitud con las cepas de referencia pertenecientes a Herbaspirillum, Azospirillum y el género Burkholderia. La capacidad promotora del crecimiento fue confirmada por al menos cinco aislamientos. Para esta bacteria, la raíz y brote mostraron resultados de crecimiento mayores en comparación con los observados en las plantas inoculadas con las cepas de referencia. Estos resultados indican que O. glumaepatula es colonizada por una muy diversa comunidad diazotrófica en la Amazonia brasileña. Se están llevando a cabo otras investigaciones para esclarecer la taxonomía de estas cepas y sus mecanismos para promover el crecimiento.

Dado el contexto sanitario provocado por la pandemia por COVID-19 durante el año 2020, el Ciclo Básico Común de la Universidad de Buenos Aires se dictó de forma virtual. La propuesta didáctica de la cátedra de Química Bruno-Di Risio se basó en el auto-aprendizaje y el auto-gestionamiento por parte del estudiante y en el rol tutorial por parte del docente. El presente trabajo relata la experiencia didáctica como resultado de este dictado virtual durante el primer cuatrimestre de 2020 en dos comisiones en las Sedes de San Isidro y Campana. Se exponen los recursos utilizados por la docente, así como el resultado académico por parte de los estudiantes ante las propuestas llevadas a cabo.

Did Safe Cycling Infrastructure Still Matter During a COVID-19 Lockdown?

The UK government introduced strict measures (including asking people to work from home and a lockdown) to slow the spread of COVID-19 by limiting people’s movement. This led to substantial reductions in traffic, making roads much safer for cyclists. This provides a unique opportunity to study the role played by safe cycling infrastructure. Many UK cities have provided cycling infrastructure to improve safety and encourage cycling. However, access to safe cycling infrastructure varies across neighbourhoods, potentially contributing to inequality. Since roads became safer due to the unprecedented reduction in traffic during the lockdown, safe cycling infrastructure may not play a significant role during this period. On the other hand, safe cycling lanes are often connected to amenities, potentially attracting cyclists even if they confer no additional safety benefit. That is, connectivity might matter more than safety. In this study, we utilised crowdsourced cycling data and regression models to examine the extent to which cycling intensity for non-commuting purposes changes with different types of cycling infrastructure in the city of Glasgow, Scotland, UK. In addition, we selected some areas with large increases in cycling intensity and examined the surrounding environments using Google Street View. Our results showed that non-commuting cycling activities increased significantly after the government interventions on both typical roads and safe cycling lanes while much higher increases were observed on safe cycling lanes than on other roads. A further analysis showed that there were large increases in cycling volumes on both typical roads and safe cycling lanes with good amenities and connectivity, highlighting the importance of these factors when building new cycling infrastructure. Since safe cycling lanes are not equally accessible to people, providing temporary cycling lanes during the pandemic considering these conditions could encourage people to cycle more, and thereby improve their health.
