The non-structural protein 13 (nsp13) of Severe Acute Respiratory Syndrome Coronavirus (SARS-CoV) is a helicase that separates double-stranded RNA (dsRNA) or DNA (dsDNA) with a 5' → 3' polarity, using the energy of nucleotide hydrolysis. We determined the minimal mechanism of helicase function by nsp13. We showed a clear unwinding lag with increasing length of the double-stranded region of the nucleic acid, suggesting the presence of intermediates in the unwinding process. To elucidate the nature of the intermediates we carried out transient kinetic analysis of the nsp13 helicase activity. We demonstrated that the enzyme unwinds nucleic acid in discrete steps of 9.3 base-pairs (bp) each, with a catalytic rate of 30 steps per second. Therefore the net unwinding rate is ~280 base-pairs per second. We also showed that nsp12, the SARS-CoV RNA-dependent RNA polymerase (RdRp), enhances (2-fold) the catalytic efficiency of nsp13 by increasing the step size of nucleic acid (RNA/RNA or DNA/DNA) unwinding. This effect is specific for SARS-CoV nsp12, as no change in nsp13 activity was observed when foot-and-mouth-disease virus RdRp was used in place of nsp12. Our data provide experimental evidence that nsp13 and nsp12 can function in a concerted manner to improve the efficiency of viral replication and enhance our understanding of nsp13 function during SARS-CoV RNA synthesis.

Sars Cov-2 has emerged as a global threat since its onset in December 2019. India has also been severely affected by the dreadful Corona Virus and is currently battling one of the worst pandemics of history. WHO and the world medical fraternity are putting their efforts to materialize a treatment or vaccine for this novel virus. A randomized open label parallel group study was designed in a Lucknow based level 2 COVID hospital to evaluate the efficacy of Ayurvedic interventions in the management of asymptomatic and mild COVID 19 patients. To evaluate the efficacy of Ayurveda in the management of mildly affected COVID-19 patients. The current trial was an open label randomized 10-day study. Total 120 asymptomatic and/or mild Covid-19 positive patients fulfilling inclusion criteria were randomly grouped into three. RT-PCR of all the patients were done on 5th, 7th and on 10th day respectively. The observations were noted and results were analyzed statistically. Kruskal-Wallis test and Wilcoxon Sign rank test were used for data analysis where applicable. Improvement in symptoms, enhancement in Agni and recovery from COVID infection was observed. The results obtained were encouraging and showed better viral clearance and control of symptom progression in the patients placed on Ayurvedic medications. The promising results in the study showed that an approach involving Ayurveda can be helpful for the management of the mild COVID-19 patients. Ayurveda can be used to limit community spread and check disease progression to a more appalling state. Trial was registered with Clinical Trials Registry- India (CTRI registration number: CTRI/2020/06/025800.

In order to support a better understanding of the current scenario of the crisis installed by the New Coronavirus (SARS-CoV-2: COVID-19) in the flower retail trade, it is presented a descriptive exploratory research carried out between April and May 2020 with 30 flower shop managers located in the Southern region of Brazil. The results showed that the most significant impacts were the drop in the number of customers, which reduced the company’s income when compared to the values sold before the pandemic, and that the pandemic scenario generated a 45.3% reduction in financial transactions on average. Among the flower shops in this survey, 70% of flower shops were closed for an average of 21.4 days, when they were reopened with restrictions on attendance related to hours or the number of people inside the stores. The e-commerce and social media were the main tools to confront this situation, and the most used Apps were WhatsApp and Instagram. But other actions such as discounting on purchases by quantity, free delivery and marketing in the surrounding were also registered. Among the possible complementary actions in order to face this crisis and which can have positive effects, it is highlighted the online courses directed to the trade of gardening kits, the adoption of the Just In Time (JIT) methodology that can result in partnerships with local producers in order to reduce inventory costs and purchase prices, and the organization of collective purchasing groups to bargain prices with wholesalers, as well as the reduction of transportation and operational costs at Veiling in Holambra.

A agricultura tem mostrado através dos tempos, responsabilidade histórica na transformação dos espaços naturais, proporcionada pela crescente necessidade de produção de alimentos. A intensa discussão dos problemas ambientais da atualidade tem exigido reflexão sobre a relação homem-natureza. O presente artigo, resultado de pesquisa de mestrado no Vale do Bananal, município de Salinas, Minas Gerais, do Programa de Pós-Graduação da Universidade Estadual de Santa Cruz, traz elementos da representação social do pequeno produtor rural, sobre meio ambiente e práticas agrícolas, mediante análise de seu discurso representado na coletividade e de sua prática agrícola de base familiar. Foi utilizada a teoria das Representações Sociais com a pesquisa de campo por meio da entrevista semi-estruturada e observação direta. A análise dos dados ocorreu através do Discurso do Sujeito Coletivo. Identificou-se vários problemas ambientais como desmatamento, uso de agrotóxicos, descaracterização da mata ciliar, erosão, contaminação de rio. Pode-se inferir que embora o pequeno produtor rural do Vale do Bananal no município de Salinas, possua uma percepção naturalista do meio ambiente, há indícios de práticas de uma agricultura sustentável.

En Centroamérica, el maíz y el frijol son usualmente cultivados por agricultores que tienen pequeñas áreas de tierra y viven en situación de vulnerabilidad. El cambio climático representa una importante amenaza para estos pequeños agricultores de granos básicos, poniendo en peligro sus sistemas de producción y medios de vida. El uso de Adaptación basada en Ecosistemas (AbE) puede ayudarlos a enfrentar el cambio climático, pero existe poca información sobre el tema. El objetivo de este trabajo fue caracterizar las estrategias de AbE que los pequeños agricultores de granos básicos de cuatro paisajes de Guatemala y Honduras utilizan para aumentar su resiliencia frente al cambio climático. A través de entrevistas y mediciones en campo en 160 fincas, entre julio 2014 y junio 2015, 1) se exploró qué tan común es el uso de las diferentes prácticas de AbE, 2) se documentaron las características biofísicas de estas prácticas, y 3) se determinó si la implementación y características de las prácticas difieren entre paisajes. Los resultados indican que muchos de los pequeños agricultores de granos básicos usan prácticas de AbE, aun cuando cultivan áreas pequeñas (parcelas de maíz de ~0,68 ha), tienen un bajo nivel de educación y bajo acceso a capacitaciones o consejos técnicos. Las prácticas más comunes fueron el uso de árboles dispersos, huertos caseros y cercas vivas. Resultados de un análisis de ANDEVA indican que hubo diferencias significativas en el uso de diferentes prácticas entre agricultores, que sugiere que las adoptan en respuesta a sus medios de vida y condiciones biofísicas. El estudio sugiere que los pequeños agricultores de granos básicos tienen la experiencia y el conocimiento para utilizar prácticas de AbE, y que ven beneficios derivados de su implementación; sin embargo, para ampliar el uso de estas prácticas, se necesita más apoyo técnico, financiero y político.

Introducción. En Centroamérica, los pequeños caficultores están amenazados por el aumento en las temperaturas y los cambios en los patrones de lluvia. Una estrategia para apoyar a las comunidades de pequeños agricultores a adaptarse al cambio climático es el uso de prácticas de Adaptación basada en Ecosistemas (AbE), las cuales integran la conservación, restauración y manejo sostenible de ecosistemas y biodiversidad. Sin embargo, existe poca información de cómo los productores utilizan estas prácticas. Objetivo. El objetivo de este estudio fue describir el uso de once prácticas de AbE entre pequeños agricultores de café, de cinco paisajes ubicados en Costa Rica, Honduras y Guatemala, así como comparar las características biofísicas y frecuencia de uso de las mismas entre paisajes. Materiales y métodos. La caracterización de las prácticas de AbE se realizó utilizando mapeo participativo, entrevistas, y mediciones de área y estructura y composición de la vegetación, entre julio 2014 y junio 2015. La comparación entre paisajes se realizó mediante un análisis de varianza. Resultados. Los resultados indican que muchos pequeños agricultores de café están implementando activamente el uso de prácticas AbE, entre ellas el uso de árboles de sombra en cafetales, los surcos en contorno, los huertos caseros y las cercas vivas. Sin embargo, existieron diferencias importantes entre paisajes en cuanto al uso y las características de AbE implementadas. Conclusión. Estas diferencias sugieren que los productores adaptan el uso de dichas prácticas en respuesta a los contextos sociales y biofísicos del medio en donde viven, y que existe valor en hacer intercambio de productores de diferentes regiones para compartir su conocimiento sobre estas prácticas.

Este artigo demonstra que o setor da pesca artesanal enfrenta dificuldades para acessar políticas públicas de crédito, o que colabora com a manutenção de mecanismos informais de financiamento. O estudo de caso realizado no Litoral Centro-Sul de Santa Catarina partiu de entrevistas semiestruturadas com pescadores e atravessadores, seguindo-se da análise de discurso e da mobilização de conceitos da sociologia econômica para entender as relações entre esses dois grupos de atores sociais. Os resultados evidenciaram que, embora a criação de uma linha de crédito específica para a pesca artesanal no quadro do Programa Nacional de Fortalecimento da Agricultura Familiar (Pronaf) tenha origem em uma reivindicação do setor, o acesso aos recursos dessa linha ainda é muito baixo. A análise apontou que os sistemas de financiamento formais e informais adotados pelos pescadores artesanais estão imbricados nas relações sociais. Os sistemas de financiamento informais têm se mantido mesmo após a criação de uma linha de crédito para a pesca artesanal. A associação de oferta de serviços financeiros com a comercialização do pescado fortalece laços assimétricos de poder entre pescadores e atravessadores. O crédito oficial é acessado por um pequeno número de pescadores artesanais, prevalecendo formas tradicionais e informais de financiamentos pouco favoráveis aos pescadores.

The article analyzes the specific public policies of rural credit and agricultural insurance for family farms, it aims to check their compatibility with two distinct technological paradigms: productivist and agroecological. On methodology, a case study was conducted at the extreme western region of Santa Catarina. The data survey was composed by interviews with 37 family farmers and 21 local operators of public policies. This region is characterized by the presence of a family farm socioeconomic dynamics. The analysis reveals a duality in public action. In one hand, policies to agroecology were created. In other hand, the operation of those public policies analyzed, prioritizes the increasing productivity of family farming units with high dependence on external inputs, while disregards the profitability per area and the enhancement of local resources. We conclude that public action has limited the adoption of agroecology as a technological paradigm.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a novel etiological agent of coronavirus disease 2019 (COVID-19). Nigella sativa, commonly known as black seed or black cumin, has been a historical and traditional plant since thousands of years. Based on their therapeutic efficacy, the chief components of terpenoids and flavonoids were selected from N. sativa seeds and seed oil. This study was designed to check the antiviral efficacy of N. sativa main phytoconstituents against five potential targets of SARS-CoV-2 using in silico structure-based virtual screening approach. Out of twenty five phytocomponents, ten components showed best binding affinity against two viral proteins viz. N-terminal RNA binding domain (NRBD; PDB ID: 6M3M) of nucleocapsid protein and papain-like protease (PL-PRO; PDB ID: 6W9C) of SARS-CoV-2 using AutoDock 4.2.6, AutoDock Vina and iGEMDOCK. PASS analyses of all ten phytocomponents using Lipinski's Rule of five showed promising results. Further, druglikeness and toxicity assessment using OSIRIS Data Warrior v5.2.1 software exhibited the feasibility of phytocomponents as drug candidates with no predicted toxicity. Molecular dynamics simulation study of NRBD of SARS-CoV-2 nucleocapsid protein-alpha-spinasterol complex and PL-PRO-cycloeucalenol complex displayed strong stability at 300 K. Both these complexes exhibited constant root mean square deviation (RMSDs) of protein side chains and Cα atoms throughout the simulation run time. Interestingly, PL-PRO and NRBD are key proteins in viral replication, host cell immune evasion and viral assembly. Thus, NRBD and PL-PRO have the potential to serve as therapeutic targets for N. sativa phytoconstituents in drug discovery process against COVID-19.
