Resultados: No Brasil, no período analisado, 25.262 casos de COVID-19 foram divulgados e 1.532 pessoas foram a óbito. Dados do IBGE de 2019 apontam cerca de 119,3 milhões de brasileiros vivendo em situação de miséria, com renda menor que um salário mínimo por mês. Além disso, segundo o Censo 2010, somente 3,8% da população brasileira tem acesso à água potável, o que pode dificultar medidas de prevenção. Conclusão: Há, diante do cenário da pandemia, a necessidade de uma maior atenção às pessoas que se encontram em vulnerabilidade social no Brasil. Escolheu-se a cultura do maracujazeiro amarelo como objeto inicial do trabalho em função da necessidade dos intensos tratos culturais nas fases de plantio, floração e colheita. Inicialmente, pesquisas de campo indicaram a seleção Afruvec como melhor material genético para plantio. Conhecendo-se a disseminação do vírus do endurecimento dos frutos (PWV), principal doença do maracujá, construiu-se uma estufa com tela anti-afídeo para produção das mudas no sistema de tubetes. Visando a transferência de tecnologia de produção, realizou-se dia de campo, reuniões e várias palestras técnicas. Durante o desenvolvimento do plantio em 2003, os produtores decidiram criar a BauruFrutas (Associação de Fruticultores de Bauru e Região), a fim facilitar o processo de comercialização e tentar obter melhor rentabilidade com a atividade. Posteriormente, foi elaborada a logomarca da Associação e com os recursos da 2a fase do projeto Fapesp, adquiriu-se linha completa de processamento de polpa para sustentabilidade da produção. Em 2005 visando dar continuidade aos trabalhos, conseguiu-se a aprovação junto ao CNPq de projeto de extensão direcionado a agregação de valor e nesse sentido, adquiriu-se equipamentos para produção da farinha da casca, utilizada na alimentação humana para redução de colesterol e diabetes e saquinhos de polpa de 100 gramas com código de barra. Visando conscientizar os produtores com o sério problema do vírus do endurecimento dos frutos do maracujazeiro, foi realizado em outubro de 2005 um seminário sobre este assunto, que resultou na publicação de um livro com apoio dos palestrantes e da Fundação de Ensino e Pesquisa (Funep) de Jaboticabal. A Associação obteve a cessão de uso de um barracão inativo junto ao Departamento de Estradas e Rodagem (DER) que foi utilizado como entreposto para classificação e armazenamento dos frutos. Middle East respiratory syndrome-related coronavirus (MERS-CoV) is responsible for sporadic infections in mostly Middle Eastern countries, with occasional transfer elsewhere. A key step in the MERS-CoV replication cycle is the fusion of the virus and host cell membranes mediated by the virus spike protein, S. The location of the fusion peptide within the MERS S protein has not been precisely mapped. We used isolated peptides and giant unilamellar vesicles (GUV) to demonstrate membrane binding for a peptide located near the N-terminus of the S2 domain. Key residues required for activity were mapped by amino acid replacement and their relevance in vitro tested by their introduction into recombinant MERS S protein expressed in mammalian cells. To aid in characterization, we developed a generalizable strategy to predict antibody-binding epitopes within individual proteins and entire proteomes. Specifically, we selected antibody-binding peptides for 273 distinct sera out of a random library and identified the peptides using next-generation sequencing. To predict antibody-binding epitopes and the antigens from which these epitopes were derived, we tiled the sequences of candidate antigens into short overlapping subsequences of length k (k-mers). We used the enrichment over background of these k-mers in the antibody-binding peptide dataset to predict antibody-binding epitopes. As a positive control, we used this approach, termed K-mer Tiling of Protein Epitopes (K-TOPE), to predict epitopes targeted by monoclonal and polyclonal antibodies of well-characterized specificity, accurately recovering their known epitopes. K-TOPE characterized a commonly targeted antigen from Rhinovirus A, predicting four epitopes recognized by antibodies present in 87% of sera (n = 250). An analysis of 2,908 proteins from 400 viral taxa that infect humans predicted seven enterovirus epitopes and five Epstein-Barr virus epitopes recognized by >30% of specimens. Analysis of Staphylococcus and Streptococcus proteomes similarly predicted 22 epitopes recognized by >30% of specimens. Twelve of these common viral and bacterial epitopes agreed with previously mapped epitopes with p-values < 0.05. Additionally, we predicted 30 HSV2-specific epitopes that were 100% specific against HSV1 in novel and previously reported antigens. Experimentally validating these candidate epitopes could help identify diagnostic biomarkers, vaccine components, and therapeutic targets. The K-TOPE approach thus provides a powerful new tool to elucidate the organisms, antigens, and epitopes targeted by human antibody repertoires. Outro tipo de levantamento analisado foi o das finalidades de usos das águas subterrâneas. Do total do cadastramento foram analisados 8.330 poços tubulares (92 %), 513 cacimbas (6%) e escavados e 148 fontes naturais (2%). Para o levantamento da qualidade da água, realizou-se, apenas análises de sólidos totais dissolvidos onde pôde-se definir que: 1.613 pontos analisados possuem água doce; 2.020 pontos analisados têm água salobra; e 2.512 pontos analisados possuem água salgada. Contata-se assim uma tendência a água salgada, esperada para a região em função das condições hidrogeológicas em que estão localizadas as captações subterrâneas. Observa-se então que há potencial para acelerar a solução dos problemas de falta de água para o abastecimento e para a agricultura familiar voltados à população rural difusa, mediante a implementação de tecnologias apropriadas.
